Season 1 General Spoilers:

01/04 - I hear the big ol' possibly one or two or twenty-eyed monster is making it's return, just in time for Feb sweeps. In Feb, another female character will disappear, a la Clare, but this one's a series regular so I'm not so worried.
Kate's backstory looks like it is good ol' fashioned crime (goes way, way back) but of course will have a new-fangled JJ Abrams' esque twist. And the secret to her past will be drudged up from the bottom of a waterfall, in a suitcase to which only the marshall has the key. Only prob? He's currently six feet under, remembah? And about the Shannon/Bonne thing: they might have to give their Family Friendly award back (you know, that one they just won). I thought he was kidding til he told me about the ep. Lordy, lordy, I need to confess just hearing about it. We'll soon get the backstory on Michael and Walt, and here's what I know: Walt's momma was a big fancy shmancy lawyer and Michael, an artist, which wasn't exactly this money and power monger's thang. It seemed HER decision not to have Michael in Walt's life and he seems most definitely a goodie in a world of baddies … including, possibly, Walt's stepdaddy. Source: Kristin on E!Online
12/27 - In the second half of the season: Sun will mysteriously go missing and the island monster will make a few reappearances. Source: SpoilerFix.com's visitor Mu
12/20 - Ian Somerhalder (Boone) will be shown kissing one of his fellow castaways in an upcoming scene on Lost. Two of the characters are related in a way that is different than what we think. One of the much loved GOOD guys -- at least, he's seemed that way so far -- is going to kill someone. Source: Kristin on E!Online
11/29 - I’m hearing that Kate and Sawyer will be more flirty-birdy than she and Jack (at least in the beginning). And that Sayid is going to get a romance of his own. I’m hearing that the tide will be coming in and that fuselage will be rolling out … They’ll all scramble to get all the goods that they can out of the wreckage before it washes away. I'm hearing Claire will not be around forever, though she will be around for a while ... Locke and Sawyer are not going to go crazy due to meds (see the 11/17 spoilers below about mind-altering medication). Source: Kristin on E!Online
11/29 - Locke makes a huge discovery. Someone will build a raft to try to escape. We'll meet other folks on the island who weren't on the plane, and learn two secrets about Hurley. The Bermuda Triangle continues to overlap with the Jack-Kate-Sawyer Triangle. (''I have a feeling that in the near term, it'll be Sawyer and Kate, and in the long term, it'll be Jack and Kate,'' says Fox. ''[But] there's not a whole lot of room for romance in the situation that these people are dealing with.'') Let's also heed this advice from Lindelof: ''The flashbacks serve as a great conduit to learn more about these characters, but that's not all they're there for. The idea that these people — way before they got on this airplane — have interacted with each other either directly or through third parties is one of the cool pieces of tapestry of the show.'' Source: Entertainment Weekly (Thanks Fabian for the tip.)
11/23 - Sayid links up romantically with one of the other women on the island. Source: About.com (Thanks Lori for the tip.)
11/17 - Someone on the show is on serious medication (mind-altering), and they are running out. And I'll rule out only one character: Vincent. Woof. Source: Kristin on E!Online
11/15 - Jack's Father's body WAS on the plane. Source: The Fuselage (Thanks irishmegs88 for the tip.)
11/14 - A cargo plane shipping zoo animals also crashed on the island once. Source: The Fuselage (Thanks irishmegs88 for the tip.)
11/12 - According to an online chat with Daniel Dae Kim, a Jin POV episode is slated for either episode 15 or 16. Source: The Fuselage (Thanks Mu for the tip.)
11/09 - Each “Lost” writer had to pull three names out of a hat. [Lost writer Paul] Dini got Locke, Sawyer and Sun and was instructed to write a history for each of the three. Dini says Locke having the same name as the philosopher is not a coincidence. “… the monster is sort of a reflection of yourself. The pilot saw it in horror and he was killed because he feared the monster. Locke saw the monster with true awe, therefore, he was able to survive his encounter,” Dini said. Though we may never see the entire monster, Dini says it may be "a Loch Ness Monster kind of thing.” The characters of Scott and Steve, introduced last week, will soon have speaking parts. At least one main character and several minor ones will die before season’s end. Source: Ain't It Cool News
11/08 - In a later episode, Sawyer and Kate stumble upon a waterfall that looks breathtakingly beautiful on the surface … but when they jump in, they find some disturbing plane wreckage, including the key to Kate’s story. Jack is going to get beaten to a bloody pulp. Source: Kristin on E!Online
11/02 - Sun's father is a leader in big business in Korea, owns a large company involved in the auto industry. A little known fact is that the reason Jin had the watch (presumably in his luggage) is that he was on a business trip to deliver the watches to his father in law's business cronies in both Australia and the U.S. where he has interests. Whether or not Sun's father is in the Korean mafia is something we will be addressing in future episodes (when you see Jin's side of the story). - From Javier Grillo-Marxuach, writer of episode 6 Source: The Fuselage (Thanks Jenn for the tip.)
